Uji Aktivitas Antibakteri Bakteriofaga HK terhadap Escherichia coli O157H7 sebagai Agen Penyebab Foodborne Disease

Abstract

Foodborne disease adalah penyakit pada manusia akibat mengonsumsi makanan yang tercemar mikroba patogen asal hewan. Salah satu penyebab foodborne disease adalah Escherichia coli (E.coli) O157H7. Alternatif terapi terhadap infeksi bakteri patogen adalah memanfaatkan isolat bakteriofaga (faga) HK sebagai antibakteri. Tujuan dari penelitian ini adalah untuk menentukan aktivitas antibakteri pada faga HK terhadap E. coli O157H7. Aktivasi faga HK terhadap E. coli O157:H7 dilakukan melalui perhitungan konsentrasi faga HK dengan teknik Double Layer Agar (DLA), uji Konsentrasi Hambat Minimal (KHM) dan Konsentrasi Bunuh Minimal (KBM) serta perhitungan Multiplicity of Infection (MOI). Pada penelitian ini dihasilkan nilai KHM dari faga HK sebesar 6,35 x 102 PFU/ml dan nilai KBM faga HK sebesar 6,35 x 103 PFU/ml sedangkan nilai MOI dari faga HK sebesar 0,9621 x 10-6 PFU/CFU. Faga HK memiliki aktivitas sebagai antibakteri terhadap E.coli O157:H7
